Abstract: Up until today, pancreatic cancer is one of the deadliest and fastest-killing cancers. A late diagnosis, the lack of effective therapies and the tumor’s chemoresistance are the principal causes of such elevated mortality rates. Pancreatic ductal adenocarcinoma (PDAC) is the most common type of pancreatic cancer and its noticeable resistance to chemotherapy drugs is favored by its dense stroma and high interstitial fluid pressure (IFP). In this work, it was our objective to design a system that allowed us to recreate in vitro the IFP to which PDAC cells are subjected and to evaluate the effect IFP has on their growth and morphology. For this purpose, we created a 3D printed system with which we were able to exert a maximum IFP of 29.3 mmHg over microfluidic 3D cultures of two PDAC cell lines (PANC-1 and PDX 354-gfp). Spheroids size, morphology, growth rate and distribution according to their area were analyzed for 3D cultures under different fluid pressure conditions. The results obtained suggested that PANC-1 cells grow better with an IFP of 29.3 mmHg but were inconclusive in regard to the PDX cell line used. Additionally, we tested the possibility of drug supply to tumorous cells grown in microchips using our 3D printed system with positive results.
